Effect of Hyperthermia on SCK Tumor Cell Survival in Cisplatinum treatment


Abstract
Effect of hyperthermia on cisplatinum treatment was studied in SCK tumor cells. We compared effect of cisplatinum treatment combined with heating on cell survival to cisplatinum or heat alone.
All SCK tumor cells were cultured in a medium of PRMI-1640. The cell cultures were incubated at 37¡É in a humidified atmosphere of 5% CO2 in 95% air. For these experiments, cisplatinum was dissolved in RPMI-1640 and diluted. All hyperthermia
treatments
were carried out in a temperature -contrlled circulating water-bath accurate to¡¾0.1¡É. After 7 days the tissue culture flasks were rinsed, fixed and stained, and the colonies were counted. Each experiment was repeated two times.
@ES The results were as follows.
@EN 1. There was significant difference of cell killing effect between cisplatinum with heating at 43¡Éand cisplatinum alone group(P<0.05). But at 41.5¡É, there was no significant difference between two groups.
2. Cell survival of heating with cisplatinum treatment was lower than that of heat alone(P<0.05).
3. The degree of survival fraction decreased with increasing heating temperatures and duration, also with increasing cisplatinum concentration.
It is concluded that a combination of heat and cisplatinum treatment reveals additive cell killing effect on SCK tumor cells than cisplatinum or heat alone.
